Intracellular pH regulation in human preimplantation embryos.
We report here that intracellular pH (pH(i)) in cleavage-stage human embryos (2-8-cell) is regulated by at least two mechanisms: the HCO(3)(-)/Cl(-) exchanger (relieves alkalosis) and the Na(+)/H(+) antiporter (relieves acidosis). The mean pH(i) of cleavage-stage embryos was 7.12 +/- 0.008 (n = 199) with little variation between different stages. Embryos demonstrated robust recovery from alkalo...
متن کاملRegulation of desmocollin transcription in mouse preimplantation embryos.
The molecular mechanisms regulating the biogenesis of the first desmosomes to form during mouse embryogenesis have been studied. A sensitive modification of a reverse transcriptase-cDNA amplification procedure has been used to detect transcripts of the desmosomal adhesive cadherin, desmocollin. Sequencing of cDNA amplification products confirmed that two splice variants, a and b, of the DSC2 ge...
متن کاملO-35: Over-Expression of XRCC1 As Potential Biomarker for Poor Prognosis in Human Preimplantation Embryos: Selection by Study of 84 Genes Involved in DNA Damage Signaling Pathways
Background: Chromosome abnormalities are associated with poor morphology and development in human preimplantation embryos, all together lead to poor outcomes. This study aimed to explore altered expression of DNA damage pathways in “poor morphological and development embryos with sever aneuploidies”. Materials and Methods: Surplus day-4 embryos of PGD cases were pooled in two groups: Poor progn...
متن کاملProgesterone receptor gene expression in preimplantation pig embryos.
OBJECTIVE It is not known whether progesterone, which plays a key role in establishing and maintaining pregnancy, acts directly on embryos or indirectly through the mother's reproductive tract. Since the physiological effects of progesterone are mediated by progesterone receptors (PR), the expression of PR during the preimplantation stages of pig embryos was determined. DESIGN AND METHODS Pre...
متن کاملExpression of connexins in human preimplantation embryos in vitro
Intercellular communication via gap junctions is required to coordinate developmental processes in the mammalian embryo. We have investigated if the connexin (Cx) isoforms known to form gap junctions in rodent preimplantation embryos are also expressed in human embryos, with the aim of identifying species differences in communication patterns in early development. Using a combination of polyA P...
